Comedy... Fruits Basket is popular and easy to recommend. Everyone, including myself, seems to like it to one degree or another. As for straight humor without a lot of drama, Pretty Face is probably the funniest manga I've read. I'm much more into anime than manga, though, so I haven't read as much as several of the other members here.

For comedy I'd recommend Hayate the Combat Butler. It is very funny, and if you are already a fan of anime/manga and gaming it is also very referential to that culture for tons of jokes that may be missed by others.
If you can find a copy of the Azumanga Daioh collection it is very amusing. It is a comic strip style collection not a typical "graphic novel" style manga, but still, very funny (as is the anime.)
Also, for a romantic comedy I'd go with Love*Com which is a very funny read as well as sweet.
For action, fantasy and comedy all mixed together (along with a bit of romance and a harem) I'd recommend Rosario Vampire. It starts off a little bit slower, but currently up to volume 7 is released and it is definitely picking up steam.
Another good fantasy/comedy story is Gakuen Alice. It is a great read and has enough mystery to keep it interesting. Don't let the fact that it is about elementary school kids turn you off it.
Bleach is a really good choice for some good action and the story (unlike the large amount of fillers that appear in the anime) moves along at a pretty rapid pace after the first few volumes. Tite Kubo's first manga Zombie Powder is also decent, but, though it technically "finished" after 4 volumes there is obviously a lot more story left to tell (I believe it was cancelled due to not being popular.)
There are also a ton of great romantic comedies I could mention that tend more toward the typical shojo drama at times, but I won't throw them out there in case you aren't interested.

Found another couple when going through my shelf of manga that I've read.
Kamikaze has a ton of good action, but is among the bloodier/gorier manga that I've read. I haven't read Claymore, but I've seen much of the anime so if the manga has similar blood/gore when compared to the anime, I would say they are probably at similar levels.
For a decent romantic comedy, with a decent amount of mystery as things progress (and plenty of drama as well) Chibi Vampire is a good choice. The final volume (14) is also coming out in not too long I think, so you won't have to wait forever to complete the story.
I would also agree with Oh My Goddess as a great romantic comedy. It is generally just ridiculously sweet and (what I've read of it) never hits the drama too hard (unlike some other manga I could mention.)
Also agree with Fruits Basket. Hits hard on the drama at times but is more often than not just funny and sweet.
I've only read the first 2 volumes so far but D. Gray-Man is a decent read. Not spectacular on the action front (I'd recommend Bleach long before this) but good (your opinion may vary.)
Get Backers is another possibility on the action/comedy front. I've read only the first volume, but it was a good read and had some great artwork in it.
Also, if you wanted to go for a novel, I'd say try picking up the Vampire Hunter D novels. Tons of awesome action in those books (just without the help of pictures (well...for the most part anyway.))

Video Girl Ai Fantasy, comedy and some romance too. Gunslinger Girl Even though not finished, it is still a great thought provoking sci fi/action series. School Rumble & Azumanga Daioh Both are funny high school mangas. If you aren't afraid of something in more mature humor, High School Girls & Pretty Face are hilarious. Maison Ikkoku is classic Rumiko Takahashi, very funny and satisfiying. Claymore is one of the best in fantasy. Not for the squeamish. Battle Angel Alita & B.A.A. Last Order really good sci fi adventure. Midori Days is one of the funniest mangas I've read. Oh! My Goddess There is a reason this has been one of the most loved series in Japan for over 20 years. Funny and creative. It's nice to have story a where the main characters stay true to themselves. Keiichi and Belldandy's relationship will always be the one constant in the universe. Gunsmith Cats Lots of action both with cars and guns. Expertly drawn and technically accurate. Some adult subject matter.

One thing to keep in mind with Bleach is that it is a long (and still being produced) series to get into. Currently 27 volumes have been released in English (with a new one about every 2 months) and I think they are up to something like volume 38 in Japan. Your best bet (financially speaking) is to grab the Bleach Graphic Novel box set (available here: http://www.rightstuf.com/cgi-bin/catalogmgr/NRKNRir5e4bjUrbaVf/browse/item/80300/4/0/0 ) which includes volumes 1-21 in a cool box (makes me wish I didn't get them individually actually.) Since it is a decent investment though (even if it saves money over buying individually in the long run) you may want to see if a local library may have the first few volumes available, or head out to a bookstore and read a little bit of the first volume to see if you like it. That way, if you don't, you won't feel like you wasted a bunch of money.

Claymore is one of the best in fantasy. Not for the squeamish. I'll second Claymore as well. It's actually a Dark Fantasy title and it's in my top 5 at number one.

Sci-fi: you can't beat some of the classics like Akira and Ghost in the Shell. Blame is a good read, considering it is a like a silent movie, the world is so unique, it is a must have for sci-fi fans. Fantasy: It's like every other book on the shelf. That's the US market right there. I agree Claymore is one of the best. Depending on your definition of what fantasy genre is, you can easily put Death Note, Naruto, Bleach in fantasy category. Try Mushishi, if you don't like violence with your fantasy. Action: I recently picked up Jack Frost by Yen Press and it is quite good, action wise. But if you are old enough for brutal violence, try Blade of the Immortal (warning: it makes everything else like a roller coaster ride in grandma's house... whatever that means!). Bleach and Naruto is definitely up in this category for sure. Comedy: I have been reading Welcome to the NHK. Again, that's pretty older teen. If you like Japanese humor, you must read Sayonara Zetsubou-Sensei. I like the anime better.

For comedy I suggest Azumanga Daioh and Yotsuba! (both are from the same author). Sci-fi/comedy I would suggest The Melancholy of Haruhi Suzumiya. Hope that helps, if I can think of some more, I will let you know
